A Study on the Ultrastructural and Functional Heterogeneities of Kupffer Cells in Rat Liver




Abstract
To evaluate the functional and structural heterogeneities of Kupffer cells in rat liver acinus, immunocytochemistry and morphometric analysis of Kupffer cells were carried out. The results are as follows : 1. Using an anti-rat macrophage
monoclonal
antibody numerical difference in total carbon - positive or negative immunoreactive cells were evaluated in the periportal and the perivenular areas. The total numbers of Ia immunopositive cells and carbon positive cells were predominant in the
periportal area. 2. In the stereological analysis of Kupffer cells there was some ultrastructural heterogeneities between the periportal and the perivenular regions. The shape of periportal cells was more flat than that of perivenular cells. The
area
fraction and numerical density of mitochondria and lysosome were not different between the periportal and the perivenular cells but the development of mitochondrial cristae was more prominent in the periportal cells. The phagocytic activity of
various
sized latex beads was greater in the periportal cells than the perivenular cells.3. To evaluate numerical difference in Ia antigen positive Kupffer cells between the periportal and the pervienular areas, immunocytochemistry was done by using
monoclonal
anti-rat Ia common determinant antibody.
The total numbers of Ia positive cells were greater in the periportal area than the perivenular area. From these results it was evident that the ultrastructural and functional heterogenetities exist in Kupffer cells and these suggest an important
role
in the unique function of Kupffer cells.
